I've always same problem with localhost: path like
http://localhost/mysite.com.br execute ok, when the path is
http://localhost/mysite.com I get a error (below).

You can't use the file extension .com in folder names in IIS. IIS treats it
as an executable and if executables are disabled, it just won't run. it's
functionally equivalent to calling for folder "mysite.exe" (which also won't
work)

Have you tried http://localhost/mysite.com/

(or not using special extensions, like .com, .exe etc. in your virtual
folder names)?
